Yeah, we need only to refer to the first page where he calls her by her first name, something commonly done by step-children. And I should know because I've always referred to my step-father by his given name, rather than calling him 'dad'. It's clear that they weren't in a romantic relationship on the 1st page, so he'd have no reason to call his genetic mother by her first name.

Also notice the panel at the bottom of page 3, the scene of his father marrying Tsubame has them both looking close, age-wise, to how they looked recently (rather than 2 decades younger). So that's more evidence against her being his biological mother.

And finally, referring to page 24, if they were truly blood-related, it wouldn't be legal for them to be married like that. And I kind of doubt they'd have as many enthusiastic supporters in attendance. But then again, it's hentai logic, so the author can do whatever.
